HMG-B1在人胃癌及周边组织中的表达研究及其克隆和序列分析

摘要 目的研究比较人胃癌及癌周边组织中高迁移率族蛋白-1(high mobility group box 1,HMG-B1)的表达量变化,克隆人肿瘤HMG-B1基因并对其进行序列分析。方法用半定量逆转录聚合酶链式反应(reverse transcript polymerase chain reaction,RT-PCR)检测HMG-B1的表达量,并同时利用内参18S rRNA对肿瘤及其周边组织的HMG-B1表达量进行比较。克隆人胃癌组织HMG-B1后,进行序列测定及分析。结果HMG-B1在肿瘤组织和癌旁组织中的表达量显著高于远端正常组织;对实验数据进行统计学分析,差异有显著性(P<0.05)。克隆出的一特异基因产物与HMG-B1预期长度678bp相符,测序结果与GenBank中报道序列同源性为89%。结论实验证实了文献报道的HMG-B1在肿瘤组织和癌旁组织中表达水平会升高,进而有可能作为肿瘤的诊断、生长、转移及预后的重要判定指标。成功获得了人胃癌组织HMG-B1基因编码序列,为进一步研究打下基础。
