摘要 目的观察重组人白细胞介素21(IL-21)对健康人外周血淋巴细胞的活化作用,检测IL-21对外周血B细胞表面抗原提呈相关分子的表达及对淋巴细胞亚群的影响。方法重组人IL-21与健康人外周血单个核细胞(PBMC)共培养48h,用流式细胞技术检测T、B淋巴细胞表面CD69的表达,并与未加IL-21组进行比较;同时用流式细胞术检测B细胞表面主要组织相容性复合体Ⅰ及Ⅱ类分子(MHC-Ⅰ、MHC-Ⅱ)、CD80及CD86的表达量。细胞培养96h后检测淋巴细胞亚群的变化。结果重组人IL-21能显著提高T、B淋巴细胞表面CD69的表达量,与未加IL-21组相比差异有统计学意义(P<0.05),但却不能提高B细胞表面MHC-Ⅰ、MHC-Ⅱ、CD80及CD86的表达量。经IL-21刺激后人淋巴细胞亚群中自然杀伤细胞(NK)细胞及自然杀伤T细胞(NKT)细胞比例明显增加。结论重组人IL-21可以活化人的T、B淋巴细胞,同时IL-21刺激后人淋巴细胞亚群中自然杀伤细胞(NK)细胞及NKT细胞比例明显增加。 Objective To investigate the activation of T and B lymphocytes induced by recombinant human interleukin-21 (rh IL-21) and the effect of rhIL-21 on the lymphocyte subsets of human peripheral blood lymphocytes. Methods Peripheral blood mononculear cells (PBMCs) from healthy donors were cultured in the presence of rhIL-21 for 48 hours. T and B lymphocytes were analyzed for CD3/CD19/CD69. B lymphocytes were analyzed for CD19/CD86, CD19/CD80, major histocompatibility complex (MHC)-Ⅱ and MHC-Ⅰ. Lymphocyte subsets were analyzed after the cells were cultured for 96 hours. Results IL-21 promoted a strong up-regulation of CD69 on human T and B lymphocytes (P〈0. 05). The surface molecules as surrogates for antigen presenting cell (APC) function on human B cells were not remarkably changed (P〉0.05). IL-21 increased the percentage of NK cells and NKT cells in the lymphocyte subsets. Conclusion rh IL-21 could activate the peripheral blood T and B lymphocytes, but could not promote up-regulation of surface molecules as surrogates for APC function on human B cells. IL-21 could increase the percentage of NK cells and NKT cells in the lymphocyte subsets.
