摘要
【目的】测定不同年龄犬红细胞刺激淋巴细胞增殖的能力,探索红细胞的免疫功能及其实现方式。【方法】无菌采取1~3月龄幼犬及18~20月龄成年犬的犬肝素抗凝血,分离红细胞,制备5.0×10^7mL。红细胞悬液,接种在24孔细胞培养板中培养56h后,收集红细胞上清;采用淋巴细胞转化增殖试验,分析了红细胞培养上清液单独及协同刀豆蛋白A(ConA)对淋巴细胞转化增殖的影响。【结果】无论自体或是异体,幼犬的红细胞培养上清对淋巴细胞的刺激效果均明显优于成年犬(P〈0.05);无论幼犬还是成年犬的红细胞培养上清,其自体刺激淋巴细胞增殖的效果均明显优于异体(P〈0.05)。【结论】幼体红细胞参与T细胞免疫调控的作用强于成年个体,这对于幼体免疫系统的快速发育成熟非常有益。
[Objective] The study was to understand whether erythrocytes from infant dogs and adults dogs have the same ability to promote lymphocytes proliferation. [Method] Erythrocytes from different dogs were cultured for 56hs and their supernatants were obtained. Then,these supernatants were added into canine lymphocyte cultural systems,and the lymphocyte proliferation effects were detected by MTT essay. [Result] The results show that effects of erythrocyte cultural supernatants from infancy dogs on the promotion of lymphocytes proliferation;were markedly better than those from adult ones. For cultural supernatants of erythrocyte and cooperating with mitogen(ConA), the results were also consistent. Moreover,on the efficacy of activation lymphocytes proliferation,autologous canine erythrocyte cultural supernatants were more efficient than those from heterology dogs. [Conclusion] The results suggest that erythrocytes have the ability to promote lymphocytes to liferation,and the ability can change with aging.
