浓缩铀诱发培养免疫细胞凋亡的电镜观察及CHX和ActD的防护作用

摘要 对浓缩铀235U内照射人淋巴细胞白血病细胞株Molt-4细胞和巨噬细胞株Ana-1细胞诱发的细胞凋亡及其防护因子进行了研究。实验中估算了235U在不同阶段培养细胞中的辐射累积吸收剂量。通过透射电镜的形态观察,发现Molt-4和Ana-1两株免疫细胞在受235U内照射作用下,可诱发核断裂,核染质边聚,以及呈现膜包裹着的凋亡小体形成为特征的细胞凋亡。而蛋白质合成抑制剂CHX和RNA合成抑制剂ActD可显著抑制由核素235U诱发免疫细胞DNA链断裂的作用,从而呈现出明显的对235U内照射诱发免疫细胞凋亡的防护作用。 The apoptosis in Molt-4 cell (a human acute lymphoblastic leukemia cell line)and Ana-1 cell(a macrophage cell line) were studied after internal irradiation with enriched uranium 235U.The cumulative radiation absorption dose of 235U in cultural cells in different periods were calculated.The morphological changes which were observed by electron microscope indicated that Molt-4 and Ana-1 immune cells after incubation with 235U,displayed chromatin fragmentation,margination of condensed chromatin,as well as the membrane-bounded apoptotic body formation.The finding shows that 235U can significantly increase the DNA fragmentation in immune cells,which may be inhibited statistically by CHX and ActD.So both CHX and ActD show radioprotective effect on Molt-4 and Ana-1 cells irradiated by 235U.
