大黄对失血性休克大鼠小肠粘膜组织中一氧化氮合酶和内皮素表达的影响 被引量:1

Expression of nitric oxide synthase and endothelin-1 in small intestinal mucosa in haemorrhagic shocked rats and affection of rhubarb on them

摘要 目的:研究大黄对失血性休克大鼠再灌注后小肠粘膜组织中诱导型一氧化氮合酶(iNOS)和内皮素(ET)表达的影响。方法:雄性SD大鼠36只随机分成对照组(不进行休克处理)、模型组(再灌注前胃管注入生理盐水10ml/kg)、大黄组(再灌注前胃管注入大黄液,大黄20g/kg)。复制失血性休克及复苏大鼠模型,大黄于术前胃管注入,造模完成后观察各组大鼠小肠粘膜病理学变化,用免疫组化法检测肠粘膜组织iNOS和ET的表达。结果:肠粘膜损伤程度分级以对照组最轻,模型组最重,大黄组居中,各组间差异有统计学意义(P<0.05)。对照组、模型组、大黄组Chiu′s评分分别为(0.33±0.78)、(3.63±0.74)、(0.97±0.69)分,模型组和大黄组明显高于对照组(q值分别为15.4986、3.0058,P<0.05),大黄组明显低于模型组(q=12.4928,P<0.05)。模型组iNOS表达明显高于对照组、大黄组(q值分别为4.1143、3.4482,P<0.05),大黄组比对照组有增高趋势,但两组间差异无统计学意义(P>0.05)。模型组和大黄组ET表达水平高于对照组(q值分别为4.7982、4.1984,P<0.05),与模型组相比,大黄组肠粘膜ET表达虽有所降低,但差异无统计学意义(P>0.05)。结论:大黄能抑制失血性休克再灌注小肠粘膜组织iNOS的表达而起到保护肠粘膜的作用。 Objective: To observe the expressions of inducible nitric oxide synthase (iNOS) and endothelin-1 (ET-1) in the small intestinal mucosa of hemorrhagic shock rats after reperfusion and affection of rhubarb on them. Methods: Thirty-six male SD rats were randomly divided into three groups, Control group, without shock; model group, before reperfusion 10 ml/kg saline stomach injectiong rhubarb group, reperfusion stomach before injection of rhubarb, rhubarb 20 g/kg. Copy hemorrhagic shock and resuscitation rats, Rhubarb in the preoperative stomach implantation, model after completion of treatment of small intestinal pathology changes, immunohistochemical detection of intestinal mucosa of inducible expression of iNOS and ET. Results: Mucosa damage to the control group graded the lightest, most model group, the middle group of rhubarb, Each group had a significant difference (P 〈0.05). Chiu's score control group, model group, rhubarb group (0.33±0.78) (3. 63±0. 74), (0.97±0.69), and rhubarb model group was significantly higher (P 〈0.05). Rhubarb group was significantly lower than the model group (P 〈0.05). Model group iNOS expression was significantly higher than the other two groups (P 〈0. 05), rhubarb group than the control group increased trend, but the difference between the two groups was not significant (P 〉0. 05). Rhubarb model group and the level of expression of the ET control group (P 〈0.05), compared with the model group, rhubarb group, although the expression of intestinal ET lower, but the difference was not statistically significant (P 〉0.05). Conclusions: Rhubarb can inhibit hemorrhagic shock and reperfusion of the small intestinal mucosa iNOS expression organizations play the role of intestinal mucosal protection.
