摘要
[目的]为猪的分子育种研究提供理论依据。[方法]采用酚-氯仿法从野猪、民猪、大白猪、长白猪、杜洛克和杂种猪的耳组织中提取基因组DNA,根据GenBank上猪ACSL4基因cDNA全序列设计3对引物,进行PCR-SSCP和测序,分析ACSL4基因3UTR区的多态性。[结果]3对引物中,仅K3引物的PCR-SSCP产物有多态性,检测到3种基因型(AA、BB和AB)。在ACSL4基因3′UTR区3862 bp处发生T→C碱基突变。仅在大白猪、长白猪,杜洛克和杂种猪中发现多态性位点,而在民猪和野猪中未发现。杜洛克、大白猪和杂种猪间不同基因型的分布都有极显著差异(P<0.01),长白猪和大白猪各基因型的分布无显著差异(P>0.05)。[结论]该研究为丰富猪种的种质特性资料库和充分利用种质资源提供了依据。
[ Objective ] The aim of the research was to provide the theoretical basis for porcine molecular breeding research. [ Method] The genomic DNA was extracted from ear tissues of wild pig, Min pig, Yorkshire, Landace, Duroc and their hybrids by using phenol-chloroform method. Three pairs of primers were designed according to total eDNA sequences of porcine ACSL4 gone on GenBank website to make PCR-SSCP and sequencing. And the polymorphisra of the 3'UTR region in ACSL4 gene was analyzed. [Result] Among 3 pahs of primers, only the PCR-SSCP prncucts of K3 primer had polymor- phism and 3 kinds of genotypes (AA, BB and AB) were detected. T→C base mutation occurred at 3 862 bp in the 3'UTR region of ACSL4 gene. Polymorphie sites were found only in Yorkshire, Landrace, Duroc and their hybrids, but that were not found in Min pig and wild pig. The distribution of different genotypes had the extremely significant difference among Duroc, Yorkshire and hybrid pig (P〈0.01), that between Landrace and Yorkshire had not significant difference (P 〉 0.05). [ Conclusion] This research provided basis for enriching the gemrplasm characteristic database of pig breeds and making the best of the germplasm resources.
