摘要
目的研究鼠蛛网膜下腔出血(SAH)后柔脑膜纤维化程度的检测指标。方法采用枕大池两次注血法建立动物模型。第21天取全脑、冰冻切片、MASSON染色后,观察柔脑膜胶原纤维,并用图像分析系统测量柔脑膜的灰度值及厚度,用"100×厚度/灰度值"衡量每个视野柔脑膜的胶原纤维含量;取脑脊液检测Ⅰ型前胶原羧基端肽(PICP)、Ⅲ型前胶原氨基端肽(PⅢNP)。结果①实验组柔脑膜的胶原纤维含量显著高于对照组(P≤0.01);②实验组脑脊液的PICP、PⅢNP均较对照组明显升高(P≤0.01);并均与柔脑膜胶原纤维衡量指标呈显著正相关(P≤0.05)。结论MASSON染色显示柔脑膜胶原纤维的含量,反映了柔脑膜的纤维化程度;脑脊液中的PICP、PⅢNP也间接反映柔脑膜的纤维化程度。
Objective To research the index of leptomeningeal fibrosis after subarachnoid hemorrhage ( SAH ) on experimental models of rats, Methods Rats were used to construct animal models of SAH by twice injecting autoblood solution into the cisterna magna, At the 21st day, Masson staining was performed to the frozen sections of the brains, Using multimedia chromatic colour analytical system, we did quantitative analysis towards the leptomeningeal collagen fiber, and regarded "100 × thickness/gray scale" as the index of leptomeningeal collagen fiber in every visual field, The levels of PICP, P Ⅲ NP in collected CSF were measured respectively, Results ①leptomeningeal fibrosis of experimental group increased significantly ( p ≤0. 01 ) ; ②The levels of PICP, P Ⅲ NP in CSF of experimental group rose statistically significantly , compared with control levels(P≤0.01);There was a positive correlation between PICP, P Ⅲ NP and index of leptomeningeal collagen fiber in experimental group ( P≤0.05 ). Conclusion Masson staining shows that the content of collagen fibers of leptomeninges and reflects the degree of its fibrosis. PICP, P Ⅲ NP in CSF also indirectly reflects the degree of fibrosis of leptomeninges.
