摘要
为了创制小麦抗病、优质育种的重要中间材料,加速丰产、优质、综合抗逆性好的小麦新品种选育进程,以抗赤霉病小麦品种苏麦3号和优质面包小麦安农94212、扬麦158等为亲本配置杂交组合,利用覆盖苏麦3号抗赤霉病主效QTL(Qfhs.ndsu-3BS)的4个SSR标记和优质高分子量麦谷蛋白亚基5+10特异分子标记在杂交F3代进行标记辅助选择,F4代选育到纯合的含有Qfhs.ndsu-3BS和5+10亚基的聚合体2个(J3316-8和J3316-10)。经田间赤霉病抗性鉴定,J3316-8自交后代的8个株系赤霉病抗性表现优于苏麦3号,J3316-10自交后代的2个株系赤霉病抗性与苏麦3号相当。两个聚合体农艺性状比苏麦3号有明显的改良,加之它们含有优质面包小麦高分子量麦谷蛋白亚基,因此可以作为小麦抗病和优质育种的中间材料。本研究结果进一步证明,将现代分子标记技术与传统育种方法相结合可以大大提高育种效率。
Wheat scab is a destructive disease of wheat.The incoordinate of high yield,good quality and high resistance has been the main problem in wheat breeding for scab resistance,The purpose of research is to develop parental materials and accelerate the process for breeding new varieties with high yield, good quality and high resistance.The scab resistant wheat variety Sumai 3 was crossed to Annong 94212 or Yangmai 158, which are good for bread-making quality. Four simple sequence repeats (SSR) markers flanking the major QTL (Qfhs. ndsu-3BS ) from Sumai 3 and two specific mark ers for high molecular weight (HMW) glutein subunit 1Dx5+1Dy10 were used for marker assisted selection (MAS). Two pyramided lines with both homozygous fifths, ndsu-3BS and 1Dx5+Dy10 were identified. Field identification of scab resistance indicated that 8 progenies of line J3316-8 showed better scab resistance than Sumai 3, and 2 progenies of line J3316-10 showed similar scab resistance as Sumai 3. These lines also showed improved agronomic traits than Sumai 3 and contain the good bread- making HMW-GS, 1DxS+1Dy10 , hence could be good parental materials for wheat breeding. These results further proved that the combination of MAS and traditional breeding methods would greatly enhance the breeding efficiency.
