摘要
转录因子(TF)对真核生物的转录调控有着举足轻重的作用,而这些转录调控大多不是一个转录因子可以独立完成的,还需要其他转录因子的帮助来共同完成对基因的转录控制。我们提出了一种新的基于序列的方法来预测转录因子的相互作用,利用了转录因子结合位点(TFBS)的距离,方向和相位等信息,利用酵母菌的芯片数据来实现这一思想,获得了165对统计上有相互作用的转录因子,通过与SGD数据库中的蛋白质-蛋白质相互作用的数据进行比较,可以达到约95.5%的正确率。最后我们还利用预测的结果讨论了相互作用的转录因子对在基因组上存在的一些特征,例如,他们对距离,方向,相位的倾向性。希望我们的预测有助于更好的理解真核生物的转录控制网络。
Transcription factor, has the pivotal function to the eukaryotic gene regulation, but these regulation mostly is not transcription factor to be allowed independently to complete, needs other transcription factor cooperation to form the protein compound to complete gene regulation, We proposed a sequence - based method to predict transcription factor interaction, and have used the genome sequence information, for example, the distance, the direction and the phase between the TFBS. We used the yeast chip data to confirm this method, and 165 significant interactions were identified. These included more than 95% of the known protein - protein inter- actions. We also discussed in genome existence some characteristic using the predict result, for example, affects transcription factors to the distance, the direction, the phase has the certain tendentiousness. Our prediction and characterization of TF interactions may help to understand the eukaryotic regulatory network.
