摘要
目的:探讨大蒜素对人胆管癌FRH-0201细胞凋亡的影响。方法:MTT法检测大蒜素对FRH-0201细胞生长的影响;光镜和电镜观察FRH-0201细胞形态变化;采用凝胶电泳法、TUNEL法和FCM检测FRH-0201细胞凋亡情况。结果:大蒜素能明显抑制FRH-0201细胞的生长,与大蒜素的浓度有关;光镜和电镜观察到FRH-0201细胞发生凋亡;凝胶电泳示实验组DNA梯带(DNA lad-der)形成;TUNEL检测实验组细胞与对照组细胞凋亡记数比较有显著差异(P<0.05);FCM显示,实验组可发现明显的凋亡峰,而对照组则无凋亡峰出现。结论:大蒜素在体外可诱导胆管癌细胞凋亡。
Objective :To study the effect of allitridi on apoptosis in human cholangiocarcinoma FRH- 0201 cells. Method: The growth inhibition of FRH -0201 cells were estimated using MTT assay; the morphological changes of the FRH -0201 cells were observed under light and electron microscopes; apoptosis of FRH-0201 cells was detected by DNA extraction and electrophoresis; apoptosis of FRH- 0201 Cells was examined with TUNEL method and FCM. Result: The growth of FRH -0201 ceils is inhibited obviously by allitridi and the inhibiting effect is dependent on the concentration of allitridi; apoptosis cells were observed by light and electron microscopes; the typical ladder pattern fragmention were observed by DNA gel electro- phoresis; the numbers of apoptosis cell were significant difference between experiment group and control groups with TUNEL method;Vmax of cell apoptosis was seen experimental group but not in the control by FCM. Conclusion: Allitridi can induce apoptosis in human cholangiocarcinoma FRH -0201 cells in vitro.
