摘要 目的探讨猪苓多糖(PPS)对慢性乙型肝炎(CHB)患者外周血单核细胞源树突状细胞(MoDC)的影响。方法从CHB患者外周血中分离出单核细胞(Mo),然后将Mo与GM-CSF、IL-4体外培养9天,并于培养第6天加入PPS共同培养,在倒置显微镜下观察DC的形态,用MTT法测定DC刺激同种异体T细胞增殖的能力,用ELISA检测DC培养上清中IL-12p40+p70的含量,并与对照组比较。结果PPS可刺激DC分泌IL-12p40+p70,增强DC刺激同种异体T细胞增殖的能力。结论PPS可以增强CHB患者外周血MoDC的免疫功能,提示其可能通过调节DC的功能参与免疫应答。 Objective To observe the stimulatory effect of Polyporus polysaccharide(PPS)on the immune function of monocyte-derived dendritic cells(MoDCs)from peripheral blood of patients with chronic hepatitis B.Methods Monocytes were purified from peripheral blood of patients with chronic hepatitis B with incubation of granulocyte/macrophage colony stimulating factor(GM-CSF),interleukin-4(IL-4)and PPS which was added on day 6.MoDC's morphology was observed under inverted microscope.The abilities to stimulate the proliferation of allogenic T cells by MoDCs were evaluated by MTT assay,and the levels of IL-12 p40+p70 in culture supernatant of MoDCs were detected by ELISA.Results PPS can stimulate MoDCs to secrete IL-12 p40+p70,and PPS activated DCs can enhance the proliferation of allogenic T cells.Conclusions PPS can enhance the immune function of MoDC from peripheral blood of patients with chronic hepatitis B,suggesting that PPS possibly participate in the immune response through modulation of function of DCs.
