摘要 目的:建立牛奶中氯霉素(CAP)的GC-ECD分析方法。方法:牛奶中CAP经乙酸乙酯提取,用正己烷去除脂肪,硅胶柱纯化样品,BSTFA-TMCS试剂硅烷化衍生,用甲苯溶解后进GC-ECD分析。结果:实验结果表明,在线性范围0.005-2.5μg/ml内,相关系数r=0.9981,回归方程Y=36967.77X+249.02,回收率为85.8%-94.5%,变异系数(CV)为1.7%-2.2%,最低检测限为1.0μg/kg。结论:本方法线性关系良好,回收率高,变异系数小,最低检测浓度低,应用本方法检测牛奶中氯霉素的残留量,基本上满足了灵敏、快速、准确可靠的分析方法要求。 Objective: To establish a method for the determination of chloramphenicol (CAP) in milk by GC - ECD analysis. Methods:CAP in milk was determined in toluene solution after being extracted by ethyl acetate. The fat was removed by n - hexane and the milk powder purified by silica gel column and silanized by BSTFA -TMCS. Results:The results showed that in the linear range of 0.005 to 2. 5 μg/ml, the correlation coefficient r = 0. 9981, the regression equation was Y = 36967.77X + 249.02, the recovery was between 85.8% and 94. 5%, while the coefficient of variation (CV) between 1.7% - 2. 2%, the lowest detection limit was 1.0 μg/kg. Conclusion:With reliable producitivity, a wide linear range with good recovery, RSD, and a preferable low detection limit, the result revealed that this method is sensitive, rapid, accurate.
