摘要
为研究实验性变态反应性神经炎(Experimentalalergicneuritis,ENA)和格林-巴利综合征(GBS)的发病机理,寻找GBS的有效诊断、治疗方法,本实验采用反复超速离心、稀盐酸等电点沉淀、CM-22弱酸性阳离子交换层析等方法,从牛硬脊膜内马尾神经根中提取到高纯度髓鞘碱性蛋白Ⅱ(MyelinBasicProteinⅡ,P2)。鉴定结果表明,该蛋白的分子量为15000,pl=9.5,在SDS-PAGE垂直板电泳、PAGE圆盘电泳中显示一条蛋白区带,在双向免疫扩散、微量免疫电泳中,与进口标准抗牛P2血清之间形成一条沉淀线,HPLC测定氨基酸成份与Kad-lubowski报告的国外四个实验室测定的结果大致相同,具有诱导EAN的生物活性。
To study the pathogenesis of experimental allergic neuritis (ENA)and Guillain Barre syndrome(GBS)and to find the effective methods to diagnose and treat GBS,myelin basic protein Ⅱ(P2)was isolated and purified from bovine intradural nervi coccygeus root by the acid extract、ultracentrifuge and exclusion chromatography with CM 22 column Results showed that the molecular weight was 15 000, and pI at pH 9 5 by isoelectric focu sing electrophoresis It migrated as a single band on SDS PAGE Double immunodiffusion and microimmunoelectrophoresis indicated the specificity of the P2 to standard P2 antiserum The amino acid composition of the P2 was the same as that of previously reported preparation by Kadlubowski et al It was highly neuritogenic to induce ENA in the rabbits
