酶催化动力学光度法测定镉(Ⅱ) 被引量:15

Determination of Cadmium(Ⅱ) by Enzymatic Kinetic Spectrophotometry

摘要 基于醇脱氢酶(ADH)催化乙醇生成乙醛的反应中镉离子对于酶的抑制作用原理,通过测定还原型烟酰胺腺嘌呤二核苷酸(NADH)吸光度的变化率得出其酶促反应速度,对应于镉离子浓度而制得标准曲线。检出限为0.02μmol/L。讨论了pH、温度和共存离子对测定的影响。本方法具有快速、灵敏、干扰少的优点。 An enzymatic method was applied to determine cadmium( Ⅱ ) based on the inhibition of cadmium( Ⅱ ) for the catalytic reaction of alcohol to form acetaldehyde by alcohol dehydrogenase( ADH). The rate of the absorbance change of nicotinamide adenine dinucleotide reduced form(NADH) was determined for calculating the velocity of the enzymatic reaction and calibration curve of cadmium ( Ⅱ ) was prepared. The detection limit was 0.02μmol/L. The effects of the pH, temperature and the coexisted ions on the determination were discussed. The method is rapid, sensitive and less interference.
