小归芍超临界提取物抑制胃癌细胞系BGC-823增殖的实验研究 被引量:2

摘要 目的探讨小归芍超临界提取物抑制胃癌细胞系人类胃癌细胞823(in human gastric carcinoma 823cell,BGC-823)增殖的作用。方法将终浓度为50、100、200 mg/L小归芍超临界提取物作用于胃癌细胞BGC-823细胞48 h,观察细胞变化并用四甲基偶氮唑蓝法分析细胞生长抑制、膜联蛋白V碘化丙啶(propidium iodide,PI)双染法检测细胞凋亡率的改变和PI法观察细胞周期的变化。结果①在一定浓度范围内,与对照组相比小归芍提取物能够明显抑制胃癌细胞BGC-823的生长,抑制率随剂量增加而增加,并能够增加细胞的凋亡率(P<0.05或P<0.01);②细胞增殖周期G1期比例明显升高(P<0.05或P<0.001)。结论小归芍超临界提取物可明显抑制胃癌细胞BGC-823的增殖,诱导凋亡以及对细胞周期G1期具有明显的阻滞作用。 Objective To study the depressant effect of the extract of Xiaoguishao on the proliferation of human gastric cancer cell line-in human gastric carcinoma 823 cell(BGC-823). Methods BGC-823 cells were treated with the supercritical fluid extract of Xiaoguishao at dose of 50,100,200 mg/L for 48 h respectively. The cell growth was observed by invert microscope and inhabitory rate was studied by methyl thiazolyl tetrazolium assay; The apoptosis and cell cycle were analyzed by flowcytometry with annexinV/ propidium iodide(H) and PI assay. Results ①Within a concentrtion coverage, BGC-823 cells proliferation were inhibited by the extract of Xiaoguishao and tha inhibitory rate on human gastric cancer cells wasincreased with the increase of dosage. The apoptotic rate of BGC-823 cells was increased by the extract of Xiaoguishao ( P 〈 0.05 or P 〈0.01). ②The percentage of G0/G1 phase in the extract groups was significantly higher than in the control group( P 〈0. 05 orP 〈0. 001). Conclusion The extract of Xiaoguishao significantly suppresses the growth of BGC-823 and induces apoptosis and cell cycle arrest in GI phase effectively.
