摘要
目的:研究中药提取物NY-3对慢性应激抑郁模型大鼠肠道神经系统的影响。方法:将40只Sprague Dawley雄性大鼠随机分为正常组、模型组、氟西汀组、中药组,每组10只。应用慢性复合应激方法造模,进行体重和摄食量的测量。采用生物化学试剂盒检测大鼠结肠组织中iNOS、NO的含量,免疫组化SP法检测结肠组织COX-2的表达。结果:模型组大鼠体重明显下降(P<0.01),NY-3组体重与模型组比较显著增加(P<0.05);与正常组比较,慢性应激使模型组大鼠结肠组织iNOS和NO含量及COX-2的表达均显著升高和上调(P<0.01);NY-3与模型组比较有统计学差异(P<0.01)。结论:NY-3可通过调节肠道神经系统,来改善抑郁状态下大鼠的消化功能。
Objective: To investigate the influence of NY-3 Formula on enteric nervous system in rat model of depression. Methods: Forty male Sprague Dawley rats were randomly allocated to four groups as follows: control, depression, fluoxetine and herb group. Depression rat model was established by using chronic mixed stimulation and measured their weight and appetite. The content of colonic iNOS, NO was established by biochemistry kits and the expression of COX-2 was detected by immunohistochemical technology. Results: The weight of depression group was declined significantly compare with control groups( P 〈0. 01 )and was increased in the herb group compare with the depression group( P 〈 0. 05 ). The levels of iNOS , NO and the expression of COX-2 increased significantly in the colon of depression model rats compared to control group ( P 〈 0. 01 ). NY-3 could decrease them ( P 〈 0.01 ). Conclusion: NY-3 can improve the digestive function caused by depression by modulating the enteric nervous system.
