摘要
目的探讨苏拉明对体外培养的人卵巢癌细胞增殖的影响。方法不同浓度苏拉明作用于人卵巢癌细胞株HO-8910PM,用四甲基偶氮唑蓝(MTT)比色法检测HO-8910PM增殖情况。结果MTT结果显示50、100、200、300、400、500、600μg/mL浓度的苏拉明作用于HO-8910PM细胞96 h的生长抑制率(G I)分别是13.1%、22.6%、35.1%、43.7%、54.3%、64.8%、70.9%,不同时间及不同浓度组吸光光度值A差异有统计学意义(P<0.01)。结论苏拉明以时间及剂量效应方式抑制体外培养的HO-8910PM增殖;提示苏拉明作为HPA抑制剂有望成为转移性卵巢癌联合化疗用药。
Objective To observe the effect of suramin on the proliferation of cultured human ovarian cancer cells in vitro. Methods After human ovarian cancer cell line HO-8910PM was exposed to the suramin (50,100,200,300,400,500,600 ug/mL) , the cell curve was drawn according to cell counting and the inhibition ratio of HO-8910PM cells was measured by MTT. Results The growth inhibition ratio was significantly different in each suramin treatment group on the 4th day. It was 13.1% , 22.6% , 35. 1% , 43.7% , 54.3% , 64.8% , 70.9% respectively. The variation of extinction rates was statistically significant both in the different-time groups and different-dose groups. Conclusions It demonstrated that suramin could inhibit the proliferation of cultured HO-8910PM cells, as time-dependent and dose-dependent manners. These findings suggest that suramin be beneficial to patients with metastatic ovarian cancer as one of the inhibitor of HPA gene.
