摘要
目的:研究人乳头瘤病毒(HPV)16型在乳腺疾病中的感染率及其与乳腺疾病的相关性。方法:收集61例肿瘤组织、29例良性增生的乳腺组织的石蜡包埋切片,并从中提取出DNA。采用聚合酶链反应(PCR)以及原位杂交(ISH)的方法,从多角度、多方面的对各种乳腺疾病中HPV-16E6的存在及其在乳腺组织中的表达进行检测。结果:用PCR检出的肿瘤组织中HPV-16E6的阳性率为65.6%(40/61),良性增生组织中的阳性率为65.2%(15/23),癌旁组织的阳性率为37.04%(10/27),乳腺癌细胞株中的阳性率为66.7%(4/6);用ISH检出的肿瘤组织中HPV-16E6的阳性率为44.3%(27/61),良性增生组织中的阳性率为44.8%(13/29),癌旁组织中未发现HPV-16E6的表达。两种方法在检验中的相同率达到72.1%。结论:HPV-16E6在乳腺癌的发生过程中起到重要的作用。
OBJECTIVE: To study human papillomavirus (HPV) type 16 infection rate and its relationship with breast diseases. METHODS: Sixty-one breast cancer tissues, 27 normal adjacent breast cancer tissues, 29 benign breast tissues and 6 breast cancer cell lines' DNA were extracted and the paraffin embedded sections were collected. By using polymerase chain reaction (PCR) and in situ hybridization (ISH) methods, the existence and expression of HPV-16 E6 were detected. RESULTS: Sixty-five point six percent (40/61) of breast cancer tissues, 65.2% (15/23) of benign tissues and 37.04% (10/27) of normal adjacent tissues and 66.7% (4/6) of breast cancer cell line tested by PCR was positive of HPV-16 E6 gene. 44.3%(27/61) of breast cancer tissues and 44.8% (13/29) of benign tissues was positive in E6 gene expression in cytoplasma shown by ISH, but none of normal adjacent tissues expressed E6 gene. The rate of consistence by two methods was 72. 1%. CONCLUSION: The presence of HPV DNA and its expression in breast cancer that oncogenic genital HPV type 16 play a certain role in the pathogenesis of breast cancer.
