摘要
瘦素是由肥胖基因(obesegene)编码、主要是脂肪组织合成并分泌的一种激素,具有调控摄食和能量消耗、抑制细胞凋亡、促进细胞增殖和血管发生的作用。瘦素与其受体在乳腺癌、结肠癌等人类肿瘤有异常高表达,本文将从瘦素及其受体的结构特点、介导的主要信号通路与其促肿瘤发生机制三方面作一综述。
Leptin, a product of the obese (ob) gene, is a multifunctional hormone produced predominantly by adipocytes. Leptin has been theorized to have several functions in regulating food intake and energy expenditure , inhibiting cell apoptosis, promoting proliferation and angiogenesis in both normal and tumor cells. The expression levels of leptin and leptin receptor (OB-R) have been shown to be significantly elevated in a variety of human cancers such as breast cancer,colon carcinoma. The purposes of this review are as follows: to describe the construction features of leptin and its receptors; to explain the principal signalling transduction pathways activated by leptin; and to review the major advances leading to the current understanding of the role of leptin in the pathogenesis of a variety of cancers.
