摘要
本研究从2003年~2006年我国东北地区收集的疑似新城疫病毒感染的鸡源病料中,分离鉴定出12株可在鸡胚成纤维细胞上产生病变的新城疫病毒,并对其进行了蚀斑纯化。应用RT-PCR方法扩增含病毒融合蛋白基因47nt~420nt片段,并进行序列测定及分析。结果表明,12株NDV分离株F蛋白裂解位点序列均为112R-R-Q-K-R-F117。重要中和位点、糖基化位点及半胱氨酸残基高度保守。与GenBank中30株NDV参考毒株F基因序列比较和分析后,发现12株NDV分离株均为基因VⅡd亚型毒株,为我国目前优势流行毒株;与不同地区及宿主来源的参考毒株比较的结果表明本次分离于东北地区的毒株没有明显宿主及地域特征。
Twelve Newcastle disease virus strains isolated from the northeast of China during 2003 to 2006 were cloned using virus plaque technique. Partial nucleotides (47 nt to 420 nt) of F gene were amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) and sequenced, The results showed that the F gene cleavage sites of twelve isolates carried the same motif ^112R-R-Q-K-R-F^117. Neutralizing epitopes, glycosylation site and cysteine residues were highly conserved. Phylogenetic tree based on partial sequences of F genes from twelve NDV isolates and 30 reference NDV strains showed that all twelve isolates belonged to VIId subgenotype. No host-specific or region-specific characteristics were found among the twelve NDV isolates comparing to NDV strains isolated from distinct regions.
