人肝纤维化组织中TGF-β/Smads家族细胞转导信号通路的研究 被引量:4

摘要 目的研究慢性乙型肝炎肝纤维化组织中转化生长因子β1(TGF-β1)、Smad2、Smad4、Smad7的表达,探讨Smads在肝纤维化发生、发展过程中的作用机制。方法对32例慢性乙型肝炎患者肝穿刺标本进行病理分期,应用实时荧光定量PCR方法检测肝纤维化组织中TGF-β1、Smad2、Smad4、Smad7 mRNA表达及变化规律。结果随着肝纤维化程度加重,TGF-β1、Smad2、Smad4 mRNA的表达逐渐增加,在S4期增加最明显(P<0.01)。Smad7 mRNA在肝纤维化初期表达增加,随着肝纤维化程度加重,表达呈下降趋势,各期之间比较差异有统计学意义(P<0.05)。结论TGF-β1和Smads分子均参与肝纤维化的发生、发展,不同类型的Smads分子作用各异。 Objective To investigate the expressions of TGF-β1, Smad2, Smad4, and Smad7 in hepatic fibrotic tissues of patients with chronic hepatitis B, and the mechanism of Smads on the development of hepatic fibrosis. Methods 32 liver biopsy samples of patients with hepatitis B were obtained and pathologically classified, and real-time PCR was used to detect the expressions and changes of TGFβ1, Smad2, Smad4, Smad7 mRNA. Results With the progression of hepatic fibrosis, the expressions of TGF-β1, Smad2 and Smad4 mRNA increased gradually, and the increases were most significant at stage S4 (P〈0.01). The expressions of Smad7 mRNA were increased at the initial stage but showed a declining tendency with the development of hepatic fibrosis, and significant difference was observed between different pathologically classified stages (P〈0.05). Conclusion TGF-β1 and Smads both participate in the progression of hepatic fibrosis, and different types of Smads play different roles in it.
