摘要 目的观察经典的肿瘤干细胞标记分子CD133在人NCSLC组织和肺癌细胞系中的表达情况。方法用CD133多抗经免疫组化染色观察12例不同类型NSCLC组织中CD133的表达,经免疫荧光染色观察CD133在H446、SPC-A1肺癌细胞中的表达。用AC133单抗经流式和激光共聚焦观察人NSCLC组织、肺癌细胞系中糖基化CD133抗原(AC133抗原)的免疫活性。结果CD133多抗染色发现CD133在NSCLC组织、正常肺组织、H446细胞和SPC-A1细胞中普遍表达。AC133单抗染色发现不同类型NSCLC组织均有少量AC133(+)细胞,而在H446、SPC-A1肺癌细胞及正常肺组织中未检测到AC133(+)细胞。结论不同类型NSCLC组织中有少量的AC133(+)细胞,为深入研究肺癌发生、发展的分子机制提供了新的靶点。 Objective To investigate the expression of cancer stem cell marker CD133 in human non-small cell lung cancer (NSCLC) tissues and cell lines. Methods Expression of CD133 was detected immunohistochemically in the specimens from 12 patients with NSCI.C,and was also detected by immunofluorescence staining in H446 and SPC-A1 lung cancer cell lines using CD133 polyclonal-Ab. Immunoactivity of glycosylated CD133 antigen(AC133)was further detected in aforementioned NSCLC tissues and cell lines using AC133 monoclonal Ab by FACS and laser confocal microscope. Results Extensive expression of CD133 was detected in NSCLC,nomal lung tissues, H446 cells and SPC-A1 lung cancer cells using CD133 polyclonal-Ab. Immunoactivity of AC133 could be detected in all aforementioned NSCLC tissues, but not in H446 cells and SPC-A1 lung cancer cells using AC133 monoclonal Ab. Conclusion AC133(+) cancer cells is detected in different types of NSCLC tissues, which promise to be new target for further investigating the roles of AC133(+) cancer cells in the occurrence and development of lung cancers.
