摘要
目的:探讨细胞周期蛋白G1(cyclin G1)与子宫内膜癌发生的关系。方法:运用免疫组织化学、原位杂交技术检测25例内膜癌组织cyclin G1蛋白和mRNA的表达,并与30例正常子宫内膜进行对照。结果:免疫组化结果显示,cyclin G1蛋白在正常增殖期子宫内膜上皮细胞仅有微弱表达,而在分泌期子宫内膜上皮细胞呈高表达,且定位在细胞核内,但在高中分化和低分化子宫内膜腺癌中均无表达。原位杂交结果所示,cyclin G1 mRNA的表达与蛋白水平的表达一致,但mRNA定位于细胞的胞浆中。结论:cyclin G1低表达可能与子宫内膜癌的发生有关。
Objeetive: To explore the relationship between abnormal expression of cyclin G1 and genesis of endometrial carcinoma. Methods: Immunohistochemical method was used to test the cyclin G1 protein in normal endometrium and endometrial carcinoma tissues. In situ hybridization was used to examine cyclin G1 mRNA in them. Results: Immunohistochemical staining showed that the level of cyclin G1 proteins were only low expression of normal endometrium in proliferative phase but high expression in secretory phase. Cyclin G1 proteins were localized clearly in cell nuclei, while little expression was in both well and poorly differentiated endometrial carcinoma. In situ hybridization showed that the level ofcyclin G1 mRNA was according to that of the irnmunohistochemical staining but it was localized in cytoplasm. Conclusion: The abnormal low expression of cyclin G1 which is progesterone dependent may be involved in the genesis of endometrial carcinoma.
