摘要
目的:观察红霉素对内毒素诱导急性肺损伤(ALI)大鼠模型干预阻断作用。方法:通过鼠尾静脉注射内毒素(LPS)4 mg/kg建立大鼠ALI(acute lung injury)模型。将54只月龄和体重相近的雄性SD大鼠随机分为3组:对照组(NC组)、实验组(LPS组)、治疗组(EY组),每组18只。对照组注射生理盐水,实验组注射内毒素,治疗组注射内毒素的同时,注射红霉素粉针剂50 mg/kg。采用双抗体夹心酶标免疫(enzyme linked immunosorbent as-say,ELISA)分析法测定54只大鼠血清和肺泡灌洗液中中IL-1β、IL-6、TNF-α以及IL-8和IL-10的水平。结果:治疗组与内毒素组相比,PaO2和pH值升高,PaCO2降低,肺系数(0.62±0.07vs0.96±0.13,P<0.05)、肺含水量(85.76±0.56vs82.88±0.47,P<0.05)、肺通透性指数(7.41±0.61vs12.26±0.58,P<0.05)均有明显下降,血清和肺泡灌洗液中的IL-1β、IL-6和IL-8以及TNF-α下降,而IL-10明显升高。病理学观察治疗组肺组织病变较内毒素组为轻。结论:红霉素能够抑制内毒素诱导急性肺损伤(ALI)大鼠体内白介素1β、白介素6和白介素8以及肿瘤坏死因子α的表达,起到干预阻断急性肺损伤向恶性循环发展的作用。
Objective :To study the effects of erythromycin treatment to endotoxin-induced ALI( acute lung injury) in rat model. Methods: The rat model of ALI was made by injecting lipopolysaccharide ( LPS ) with a dose of 4mg/kg into vena caudalis. 54 male SD rats were randomly divided into three groups: control group( NC), experiment group(LPS) and treatment group ( EY), 18 rats every group. In control group physiologic saline was injected, in experiment group LPS was injected, in treatment group LPS was injected as time as erythromycin injectable powder intraperitoneal injection( 50 mg/kg). Measured the levels of IL-1β, IL-6, TNF-α, IL-8, IL-10 in serum and alveolar wash liquid of 54 rats by enzyme-linked immunosorbent assay (ELISA). Results:Compare treatment group with experiment group, PaO2 and pH stepped up, PaCO2 stepped down, lung coefficient was(0.62 ± 0.07 vs 0. 96 ± 0. 13 ,P 〈 0.05 ), lung water content( 85.76 ± 0.56 vs 82.88 ± 0.47 ,P 〈 0.05 ), lung permeability index(7.41 ± 0.61 vs 12.26 ± 0.58,P 〈 0.05 ), all decreased significantly, IL-16, IL-6, IL-8 and TNF-α in serum and alveolar wash liquid stepped down, however, IL-10 stepped up significantly. The lesion of lung tissue in treatment group was lighter than that in experiment group through pathology observing. Conclusion: Erythromycin caninhibit the expression of IL-1β, IL-6 IL-8 and TNF-α, and made a role of intervening and preventing the worse development from the acute lung injury.
