摘要
背景:激光解析/离子化-飞行时间质谱技术为以蛋白质/肽为主要成分的中药蛋白质组学研究提供了重要的技术平台。目的:对传统中药地龙蛋白/肽成分进行蛋白质组分析。设计、时间及地点:对比实验,于2004-12/2005-10在中南大学生物科学与技术学院分子生物学实验中心完成。材料:生干地龙和炮制品(沙烫)地龙,均购于湖南长沙九芝集团芝林大药房,源产地湖南。方法:以蛋白质芯片为载体,激光解析/离子化-飞行时间质谱法获取两者所含蛋白质/肽成分信息。主要观察指标:两种地龙Mr1500~13000区间蛋白质、肽分布及其相对分子质量。结果:共获得29个蛋白质/肽相对分子质量峰,其中生干品地龙17个有意义峰,炮制品地龙获取12个峰;多个相邻蛋白质峰之间质量相差一个氨基酸残基。并且生地龙与炮地龙有多组相对分子质量分别极为相近,推测可能为相同的肽段。结论:应用激光解析/离子化-飞行时间质谱技术可以形成生品地龙及炮地龙蛋白质/肽成分质量指纹图,可作为地龙数字化质控标准,并为进一步分离、纯化及验证中药地龙功能相关活性蛋白质/肽成分打下基础。
BACKGOUND: Laser desorption/ionization time-of-flight mass spectrometry technique provides important technique platform for Chinese medicine proteomics with protein/peptide as main components. OBJECTIVE: To analyze the traditional Chinese medicine Pheretima asiatica proteome by means of proteomics. DESIGN, TIME AND SETTING: The comparative trial was performed at the Experimental Center of Molecular Biology between December 2004 and October 2005. MATERIALS: Dried and processed Pheretima asiatica purchased from Hunan. METHODS: The protein/peptide from the dried and processed Pheretima asiatica was obtained, and protein/peptide information was analyzed with surface enhanced laser desorption/ionization time-of-flight mass spectrometry technique. MAIN OUTCOME MEASURES: Distribution and molecular weight of Pheretima asiatica of interval Mr 1 500-13 000 protein. RESULTS: Twenty-nine peaks of protein/peptide molecular weight were obtained. Among them, crude and dried Pheretima asiatica had 17 peaks of meaningful molecular weight, and processed Pheretima asiatica had 12 peaks. There was an amino acid residue among multiple adjacent protein peaks, and multi-groups of molecular weight of crude and processed Pheretima asiatica were extremely similar, indicating these peptides might be deduced to the same peptides. CONCLUSION: The molecular weight fingerprinting is obtained from the crude and processed Pheretima asiatica protein/peptide by laser desorption/ionization time-of-flight mass spectrometry technique; it can serve as digitalized quality and control standard of Pheretima asiatica, and provide references for further isolation, purification, and verification of protein/peptide associated with Pheretima asiatica function.
