川芎嗪预处理对大鼠心肌缺血再灌注损伤作用及其机制研究 被引量:3

摘要 目的:探讨川芎嗪预处理对大鼠心肌缺血再灌注损伤的保护作用。方法:将30只Wistar大鼠随机分为3组:假手术组、缺血再灌注损伤组、川芎嗪预处理组。建立大鼠心肌缺血再灌注损伤模型,测定SOD、GSH-Px活性,应用原位杂交技术和免疫组化S-P法检测HSP70蛋白的表达。结果:与缺血再灌注损伤组比较,川芎嗪预处理组SOD(P<0.01)、GSH-Px(P<0.05)活力显著升高,HSP70mRNA(P<0.01)及其蛋白(P<0.05)的表达显著增加。结论:SOD、GSH-Px和HSP70是心肌中重要的内源性保护物质,川芎嗪可增加SOD、GSH-Px活性,并诱导HSP70mRNA及其蛋白的表达而发挥心肌保护作用。 Objective: To probe into the protective effect of ehuanxiongzine pretreatrnent on rats' myocardial ischemia reperfusion injury. Methocks: 30 Wistar mrs were randomly divided into 3 groups: sham operation group, the group of isehemia reperfusion injury and the group of ehuanxiongzine pretreatment. The model rats with myocardial tsehemia reperfusion injury was built and ,SOD and GSH- Px activity were detected. In situ hybridization and Streptavidin-peroxidase(S- P) immunohistoehemieal rnethod were used to detect the expression of heat shock protein 70. Results: Compared with the group of ischemia reperfusion injury, the group of ehuanxionffzine pretreatment had an evident increase of vatc.lity: .SOD (P〈 0.01 ) and GSH- Px (P〈 0.05), and a remarkably increased experssion of HSP70 mRNA(P 〈 0.0 1 ) and protine(P 〈 0.05). Conclusion: SOD, GSH - Px and HSPT0 were the important endogenous protective sulxstanees in the myocardium. Chuanxiongzine can enhance the activity of SOD and GSH - Px and induee the HSP70mRNA and the expression of its protein, which was the protective effect of mycxzardium.
