摘要
目的探讨肉苁蓉提取物对败血症休克大鼠胸腺细胞凋亡的影响及其机制。方法60只大鼠随机分为实验组、空白对照组和对照组,各20只,空白对照组分离盲肠后不做结扎、穿孔处理,实验组及对照组行盲肠结扎穿孔术(CLP)建立大鼠败血症休克模型。术前14日开始实验组给予肉苁蓉溶液1.25g/kg,对照组及空白对照组给予等量0.9%氯化钠注射液。于术后12、24h取材。流式细胞仪检测各组胸腺细胞凋亡率和线粒体膜电位,分光光度法测定细胞SOD活性、MDA含量。结果实验组CLP术后12、24h胸腺细胞凋亡率均低于对照组(P<0.01),线粒体膜电位显著上升(P<0.01),细胞超氧化物转化酶(SOD)活性上升、丙二醛(MDA)含量下降与对照组比较差异均有统计学意义(P<0.01)。结论肉苁蓉提取物对败血症休克时胸腺细胞有保护作用,其机制可能是提高SOD活性,降低MDA含量,清除自由基反应,维持了细胞线粒体膜电位。
Objective To observe the effect and mechanism of extract of Roucongrong on thymus cell apoptosis in rats with septicaemia. Methods 60 rats were randomly divided into experimental group, blank control group and control group, 20 rats in each group. The rats in blank control group were not treated by hgation and perforation after cecum separation, while the rats in experimental group and control group were performed by cecum ligation perforation （CLP） to set up rat model with shock caused by septicaemia. The rats in three groups were treated by correspondent drugs 14 days before ligation （1.25 g/kg）, and the specimens were taken at 12 h and 24 h after the operation. The apoptotic rates and potential of mitochondrial membrane were detected by flow cytometer and the activity of SOD and content of MDA were determined by spectrophotometry. Results The thymus cell apoptotic rate at 12 h and 24 h after the CLP operation in experimental group was significantly lower than that of control group（ P 〈 0.01 ） ,however, the potential of mitochondrial membrane was significantly increased （ P 〈0.01）; the activity of SOD was increased, while the content of MDA was decreased, as compared with those of control group（ P 〈0.01）. Conclusion The extract of Roucongrong has protective effect on thymus cell in rats with shock due to septicaemla, its mechanism might be related with the increase of activity of SOD, decrease of content of MDA, reaction of free radical scavenging and steady of potential of mitochondrial membrane.
