人红细胞负载海藻糖后冻干保存过程的初步研究 被引量:2

摘要 目的探讨红细胞冻干长期保存的有效方法,并评价复水后红细胞各项理化指标的变化。方法设对照组(常规条件下保存的红细胞)和实验组(负载海藻糖冻干-复水后红细胞),在37℃条件下,红细胞负载海藻糖7h后,采用主要成分为含15%聚乙烯吡咯烷酮(PVP)和150mmol/L海藻糖的缓冲液作为保护液,在设定的降温程序下进行红细胞的冻干保存。冻干后置37℃的再水化液快速水化,检测各项理化指标。结果红细胞冻干再水化后红细胞和血红蛋白回收率均在80%以上,且各项理化指标与常规保存的对照红细胞间差异无显著性(P>0.05)。结论红细胞在37℃孵育7h的条件下负载海藻糖后进行冻干,复水后能保持细胞的理化稳定性和结构形态的完整性,为进一步研究长期冻干保存红细胞奠定了基础。 Objective To explore an efficient technology for long-term preservation of red blood cells and evaluate the biological activities of the RBCs after lyophilization. Methods The samples were divided into two groups:the control group (RBCs preserved in routine method), and the test group (red blood cells after freeze-drying and rehydration). Loading RBCs at 37℃ in a time span of 7 h,then put the RBCs into the protective solution which mainly composed of 150 mmol/ L trehalose, 15% polyvinylpyrrolidone (PVP) and isotonic buffer were adopted to lyophilize red blood cells at designed pro- cedure. After lyophilization,the samples were quickly rehydrated by 37℃ rehydration solution. RBCs recovery and the activity level of ATP and G-6-PD were investigated.
