保心颗粒保护阿霉素慢性心肌损伤的作用及机制研究 被引量:1

摘要 目的:观察保心颗粒对大鼠阿霉素心肌病的保护作用,并探讨其抗氧化作用机制。方法:SD大鼠随机分4组,空白对照组(A组),阿霉素组(B组,ADR组),阿霉素+保心颗粒组(C组),保心颗粒组(D组)。A组普通饲料喂养,自由饮水。B组阿霉素2.5mg/kg(用生理盐水稀释为0.5 mg/mL)腹腔注射,6天一次,共7次。普通饲料喂养,自由饮水。C组阿霉素处理同阿霉素组,药物饲料喂养(保心颗粒:普通饲料=1:20),保心颗粒组(D组):保心颗粒喂养(同前)。观察生存率、心肌谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活力;免疫组化检测CuZnSOD蛋白表达水平。结果:(1)保心颗粒可以保护阿霉素心肌病。与ADR组比较,ADR+保心颗粒组可明显提高大鼠的生存率(p<0.01)。ADR组肝、肺淤血明显,ADR+保心颗粒组则无明显淤血。(2)保心颗粒通过增加心肌抗氧化酶活力,增强心肌组织抗氧化酶mRNA转录水平以及蛋白质表达水平发挥其保护作用。结论保心颗粒从多个环节发挥其抗氧化作用,对阿霉素性心肌病具有明显的保护作用,为临床治疗心肌病提供新的思路。 Objectives: To study the protective effect ofBao-Xin Keli on cardiotoxicity induced by doxorubicin (ADR) and its anti-oxidize mechanism. Methods: Forty SD rats were randomized into four groups (n=10): control group (A), ARD group (B), ARD plus Bao-Xin Keli group( C ) and Bao-Xin Keli group( D ). Group A and B were given normal feeds, while the group B and C were given adriamycin by intraperitoneal injection at the dosage of 2.5 mg/kg. Group C and D were given Bao-Xin Keli. Survival rate of the rats and the activities of GSH-PX, CAT and SOD in myocardium in each group were detected. The expression level of Cu-Zn-SOD protein was detected by immunhistochemical method. Results: The survival rate of SD rats was significantly improved in the group treated with Bao-Xin Keli compared with that in ADR group (.p〈0.01). Bao-Xin Keli significantly enhanced the expression level of Cu-Zn-SOD protein, upregulated the activities of GSH-PX, CAT and SOD, and improved heart function when compared with control group significantly ( p〈0.01). Conclusions: Bao-Xin Keli has effect on multiple antioxidat pathways which may decrease myocardium injury in rat and protect the myocardium. It may be partly caused by reducing oxygen radicals and inhibiting oxidative stress. It is a new idea for the treatment of cardiomyopathy induced by drug.
