摘要
目的:应用人类基因芯片技术对人乳牙生理性吸收的基因表达进行分析,筛选差异表达基因,以期研究牙根吸收的基因调控机制。方法:留取与乳牙根吸收部位相对应的软组织为实验组(A)及取适量切除的牙龈瓣为对照组(B),提取总RNA,逆转录为cDNA,用Cy3和Cy5荧光染料标记,与芯片杂交后扫描。结果:有明显差异表达的共有156条,其中表达上调的基因有91条;下调的基因有65条。结论:乳牙生理性吸收可能与多个基因相关。
Objective: To screen the differentially expressed genes of physio-absorption of deciduous teeth by cDNA microarray technique for investigating the mechanism of the gene regulation of absorption during deciduous teeth. Methods : The total mRNAs were isolated from the tissues of corresponding root and gingiva and the RNAs were labeled with fluorescence Cy5 and Cy3 to prepare the cDNA probes which were then hybridized with the gene chip. Results : One hundred fifty six differential expressed genes were shown in the tissues with up-regulation in 91 and down-regulation in 65. Conclusion : There are many genes correlating to 9hysiological absorption of deciduous teeth.
