摘要
目的建立人血浆中西替利嗪浓度的HPLC测定法。方法色谱柱为Hypersil ODSC18(150mm×5.Omm,5μm);流动相为0.02mol·L^-1。磷酸二氢钾溶液(pH3.50):乙腈=77:23;流速1.0mL.min^-1;柱温30℃,检测波长230nm。结果西替利嗪血浆浓度在20~1000μg.L^-1。浓度范围内,线性关系良好(r=0.9998)。方法回收率99.54%~102.27%,日内、日间RSD均〈10%。结论本方法简单、灵敏、重现性好,适用于临床血药浓度监测.以及人体药动学的研究。
OBJECTIVE To establish an HPLC method for the determination of cetirizine in the blood plasma. METHODS The analysis was carried out by using Hypersil Cl8 column(5ttm, 150mm × 5.0mm). The mobile phase consisted of 0.02mol· L^- 1 potassium dihydrogen phosphate(pH 3.50)-acetonitrile = 77 : 23. The flow rate was 1.0mL·min^-1. The column temperature was 300C. The excitation wavelength was 230nm. RESULTS The linear range of cetirizine was 20 -- 1000μg·L^-1 (r = 0. 9998), The recoveries were 99.54 % -- 102.27 %, and RSD of intra-and inter-day were below 10 % .CONCLUSION The method is practical and accurate, and can be applicated in clinical blood medicine assay and the pharmacokinetics of human body.
