冠心病心绞痛血瘀证的血浆双向电泳-质谱研究 被引量:15

摘要 目的:进行正常人和冠心病心绞痛血瘀证患者血浆的双向电泳图谱检测,寻找心绞痛血瘀证血浆差异蛋白,探索心绞痛血瘀证的蛋白质组学特点。方法:采用双向凝胶电泳和基质辅助激光解析/电离-飞行时间质谱对6例心绞痛痰瘀互阻证患者、9例心绞痛气虚血瘀证患者和12例正常人血浆进行比较蛋白质组学研究,寻找心绞痛血瘀证血浆差异蛋白。结果:初步发现了Fibrinogenβchain、Fibrinogenγchain、α1-Antitrypsin、Hap-toglobinβchain在心绞痛痰瘀互阻证和气虚血瘀证患者中均高表达,ApoA-IV、ApoA-I在心绞痛痰瘀互阻证和气虚血瘀证患者中均中低表达。结论:初步发现Fibrinogenβchain、Fibrinogenγchain、α1-Antitrypsin、Haptoglo-binβchain、ApoA-IV、ApoA-I在心绞痛血瘀证患者和正常人血浆中存在差异表达,但结果有待运用其他生物学的方法进行具体的验证并探索其机制。 Objective : To establish 2 - dimensional gel electrophoresis images and seek differentially expressed plasma proteins of unstable angina Blood - stasis syndrome. Methods : 2 - dimensional gel electrophoresis and Matrix - assisted laser desorption/ionization time of flight mass spectrometry were used to screen the differentially expressed plasma proteins among unstable angina Blood - stasis syndrome and health volunteers. Results : Fibrinogen β chain, Fibrinogen γ chain, α1 -Antitrypsin, Haptoglobin β chain were significantly highly expressed in the plasma of unstable angina Blood - stasis syndrome patient,while ApoA- Ⅳ,ApoA- Ⅰwere decreased in the plasma of unstable angina Blood- stasis syndrome patient. Conclusion : Fibrinogen β chain, Fibrinogen γ chain, α1 - Antitrypsin, Haptoglobin β chain, ApoA - Ⅳ, ApoA- Ⅰare differentially expressed in the unstable angina Blood -stasis syndrome and control group, and the result should be validated by other biochemical technologies.
