摘要
目的分离大劣按蚊血淋巴中的丝氨酸蛋白酶并进行鉴定。方法采集感染约氏疟原虫后24h的大劣按蚊血淋巴,利用SDS-PAGE进行血淋巴蛋白的分离。然后,分别用烟草天蛾丝氨酸蛋白酶PAP1、PAP2和PAP3抗体进行Western blot,检测血淋巴中相应的丝氨酸蛋白酶。结果SDS-PAGE分离出31条血淋巴蛋白带,分子质量单位为10~200ku。Western blot结果显示,44ku蛋白能被PAP1、PAP2和PAP3抗体识别,27ku蛋白能被PAP1和PAP3抗体识别,38ku蛋白能被PAP2抗体识别。结论成功分离和鉴定出了大劣按蚊血淋巴中的丝氨酸蛋白酶。
Objective To separate and identify the serine protease in hemolymph of Anopheles dirus. Methods Hemolymph proteins of A. dirus infected with Plasrnodiurn yoelii for 24 h were collected and separated by SDS-PAGE. Then western blots were carried out with three kinds of antibody against serine protease PAP1, PAP2 and PAP3, respectively, of Manduca sexta to identify the separated proteins. Results Thirty-one bands with molecular weights of 10- 200 ku were seen in the gel of SDS-PAGE. Western blot results showed that protein with molecular weight of 44 ku could be recognised by antibodies against PAP1, PAP2 and PAP3, respectively. And protein with molecular weight of 27 ku was recognised by antibodies against PAP1 and PAP3. However, the 38 ku protein could only be recognised by antibody against PAP2. Conclusion Serine proteases in hemolymph of A. dirus were separated and identified successfully.
