摘要
目的观察中国汉族非血缘关系随机个体和家系的Rh阴性D基因完整者与C基因间的关系。方法用聚合酶链反应-序列特异性引物(PCR-SSP)扩增技术,扩增Rh血型C/E基因,D基因的外显子,检测192例RhD阳性个体,166例RhD阴性个体的血样品。结果192例RhD阳性个体,均有D基因8个外显子。在166例RhD阴性个体中,100例有ce/ce或ce/cE基因型的个体RHD外显子阴性;66例有Ce/Ce或Ce/ce基因型的个体中,34例RHD外显子完整,19例RHD外显子部分完整,13例RHD外显子阴性。结论中国人RhD阴性个体D基因完整者与C基因间可能存在联系,其在中国各民族、各地区Rh阴性个体中的发生频率和机制有待于进一步调查。
Objective To study the expression of RHC gene in serologlcally RhD negative individuals. Methods A total of 192 RhD positive samples, and 166 RhD negative samples were tested by PCR-SSP for exons of RHC/E and D genes. Results Complete expression of all the 8 exons of RHD gene were detected in all RhD positive samples. Among 166 RhD negative samples, 100 samples with ce/ce or ce/cE genotype had null RHD exons. As with the other 66 samples with Ce/Ce or Ce/ce genotype, intact RHD exons were found in 34 samples, partial D in 19, and null D in 13. Conclusion There maybe some relationship between the expression of RHC gene and the intactness of RHD gene in the RhD negative Chinese individuals. Further studies are warranted regarding the frequency and mechanisms among different populations and regions of China
