摘要
目的:探讨如何成功建立小鼠心肌梗死模型,提高动物的存活率。方法:采用结扎冠状动脉左前降支方法建立小鼠心肌梗死模型,术后4周取出心脏对心肌进行TTC和HE染色检测。结果:通过对常规手术方法进行改进,术后2周小鼠存活率由40%提高到75%。心肌颜色、心电图特异性改变及心肌TTC和HE染色证明制模成功。结论:正确使用麻醉药、顺利气管插管、准确结扎冠状动脉、减少出血与肺损伤及术后观察和护理是保证制模成功的基础。
Objective:To investigate how to establish a credible mice model of myocarclial infarction with a high survival rate.Methods:Develope mice model of myocardial infarction by ligation the left anterior descending coronary artery,and the myocardium was detected by TTC and HE staining in 2 weeks after operation.Results:Following the traditional operational method was improved,the survival rate of mice was improved from 40% to 75%.The changes of echocardiography、myocardium colour、TTC and HE staining proved the success of model.Conclusion:The factors,including correct applying of anaesthetic、favorable intubation、exact ligating of left anterior descending coronary artery、decreaseing bleeding and lung injury、observing and nursing after operation,were basis for establishing mice model with myocarclial infarction.
