摘要 目的探讨孕期持久性有机污染物(POPs)甲氧滴滴涕(MXC)暴露对孕鼠胎盘及其子代的影响。方法3月龄雌性SD大鼠40只,经适应性饲养并受孕后随机分为4组,每组10只,于孕期经腹腔注射MXC染毒,注射剂量分别为0(对照组)、16(低剂量组)、32(中剂量组)、64mg/(kg·d)(高剂量组),连续注射20d,观察MXC对孕鼠胎盘及其仔鼠的影响。结果随着MXC染毒剂量的增加,孕鼠卵巢黄体数、受精卵着床数减少,活胎数下降,不良妊娠结局增多,与对照组比较差异显著(P<0.05)。MXC中、高剂量组闭锁卵泡增多,闭锁卵泡与对照组比较呈现上升趋势(P<0.05);雄性仔鼠肛门生殖器距离(AGD)显著缩短(P<0.05),雌性仔鼠AGD未受显著影响,但其阴道开放的数量显著多于对照组(P<0.05)。结论MXC具有胚胎毒性,胎盘组织是其发挥毒性作用的靶器官之一。孕期暴露于MXC可造成胎盘滋养细胞损伤,相关细胞功能受损是其影响雌性生殖健康和子代正常生长发育的可能机制之一。 Objective To investigate the effects of exposure of gravid rats to the persistent organic pollutants (POPs), methoxychlor (MXC), on the placenta and their progeny during gestation periods. Methods According to different dosage levels of MXC [16, 32, 64 and 0 mg/(kg·d)], forty female SD rats aged 3-month were randomly divided into low-dosage group, mid-dosage group, high-dosage group and control group, with 10 animals for each. All the rats received intraperitoneal injection of MXC for 20 days. The influence of MXC on placenta of gravid rats and their progeny were observed in all aspects. Results With the increase in MXC dosage, intense changes were found in the rats, including an inerease in the number of corpus luteum of ovary, the number of nidation, fetal death and merging fetal (P〈0.05). Compared with the control group, the number of corpora atretica increased greatly both in mid-dosage group and high-dosage group (P〈0.05). The anogenital distance (AGD) of the male newborn rats was shortened significantly (P〈0.05). In female newborn rats the AGD was not affected, while the individuals with relaxed vaginal outlet increased compared to those in control group (P〈0.05). Conclusions MXC possesses embryotoxicity and the placenta is one of its target organs. Exposure to MXC during pregnancy period may injure the trophocytes of the placenta and impair the function of the cells, which may be the pathway that MXC throws its influences on the female's reproduction and affects the normal growth and development of the filial generation.
