高效液相色谱法测定犬血浆和小鼠组织、体液中的紫杉醇 被引量:11

摘要 建立了犬血浆和小鼠组织、尿和粪中紫杉醇的HPLC检测法。生物样品用乙醚提取后在C18柱上以0.01mol/L磷酸二氢钠-甲醇(4060,pH=4.3)为流动相,安定为内标,检测波长为233nm条件下进行检测。线性范围为18.7~3600μg/L,最低检测浓度为10μg/L。紫杉醇在犬血浆和小鼠肝、尿、粪中的回收率分别为86.04%~91.73%,87.62%~92.12%,86.78%~92.16%,79.83%~85.62%;日内RSD分别为2.33%~3.78%,1.37%~4.55%,3.54%~4.28%,2.30%~4.06%;日间RSD分别为3.31%~4.38%,4.72%~5.51%,6.75%~8.53%,4.43%~6.67%。用本法对家犬iv紫杉醇后的药代动力学进行了研究,并报告了有关的动力学参数。 A HPLC method was developed for the determination of taxol in dog plasma and mouse tissue,urine and feces.All samples were extracted with diethyl ether and separated on a C 18 column with the mobile phase consisted of 0 01 mol/L NaH 2PO 4 buffer-methanol(40∶60,pH=4 3).The internal standard was diazepam.The detector was set at UV 233nm.The linear range of the calibration curve was 18 7~3600 μg/L and the minimal detectable concentration was 10μg/L.The recoveries of taxol in dog plasma and mouse liver,urine,feces were 86 04%~91 73%,87 62%~92 12%,86 78%~92 16%,79 83%~85 62%,respectively.The RSDs of within-day were 2 33%~3 78%,1 37%~4 55%,3 54%~4 28%,2 30%~4 06%,and RSDs of day-to-day were 3 31%~4 38%,4 72%~5 51%,6 75%~8 53%,4 43%~6 67%,respectively. The method has been used in pharmacokinetic studies of taxol in dogs after iv 3 0 mg/kg and their pharmacokinetic parameters were reported.
