摘要
目的:探讨层粘连蛋白总糖肽(LN-Gps)增强巨噬细胞(Mφ)杀伤黑色素瘤B16-MBK细胞作用的机制。方法:将细菌脂多糖(LPS,5EU/ml),甘露聚糖(Mannan,10mg/ml),LN-Gps(200μg/ml)及PBS(对照),100μl/孔,分别刺激小鼠腹腔Mφ,并于持续刺激6,8,16,24,32,40h后观察Mφ对B16细胞的直接杀伤作用及其条件培养基对B16细胞的杀伤作用。结果:Mφ经各种刺激作用16h后,对B16细胞总的杀伤率各组无显著差异;各组在刺激后各时间里Mφ对B16细胞的直接杀伤率的动态变化相似。但各种刺激时间后收集的条件培养基对B16细胞的杀伤率动态曲线明显不同,LPS及Mannan刺激后,以6h的条件培养基杀伤作用最强(P<0.05),8h后回到对照水平,而LN-Gps组则在刺激16h后逐渐增强,40h达到高峰。结论:LN-Gps可刺激Mφ分泌一些可溶性物质杀伤B16细胞,此作用机制与其所含高甘露糖型糖链结构无关,与LPS的作用也明显不同。
Objective:To explore the mechanisms of LN Gps upregulating effect on macrophage cytotoxicity against B16 MBK cells. Methods: Macrophages, collected from BALB /c mouse peritoneal lumen, were stimulated with LPS(5 EU /ml), Mannan(10 mg /ml), LN Gps(200 μg /ml) and PBS (control) for 6,8,16,24,32,40 h and their direct cytotoxicity to B16 target cells and the conditioned media extracellular cytolysis of B16 cells were measured using MTT colorimetric methods. Results: After 16 hours' stimulation, the total cytotoxicity rates in all groups were not significantly varied, and the contacting direct cytotoxicity to B16 target cells was similar in the continuous observation time in every group. But, for the conditioned media, the effects were quite different: The conditioned media collected after 6 hours' stimulation with LPS and Mannan, showed the most potency in killing B16 cells, while with LN Gps, as the stimulation time prolonged, the cytolysis rates of the conditioned media elevated, showing the greatest increase after 40 hours' stimulation. Conclusion: LN Gps can stimulate macrophages to secrete some soluable materials in the media to kill the B16 cells, the effect is not related to the high mannose structure in the LN Gps, and is not related to LPS either.
