摘要 目的:建立大青叶水煎液血清药物化学的初步研究。方法:通过HPLC-DAD图谱和紫外光谱分析比较空白血清、含药血清和水煎液的成分异同,确定灌服大青叶后大鼠血中移行成分。结果:多次重复后,确定了处理血清的方法。大青叶给药后血中出现了3个移行成分,移行成分极性中等,其中有一成分相对含量较高,且与水煎液的HPLC图谱在相同保留时间出峰,光谱类似,其他成分可能为代谢产物。结论:血中移行成分及代谢产物将成为大青叶的体内作用基础,其来源为大青叶中的主要物质,对其进行深入的研究将为进行大青叶活性成分的进一步研究奠定基础。 OBJECTIVE To investigate the parameters of serum pharmacochemistry of Folium Isatidis. METHODS Based on the HPLC DAD chromatogram and UV spectrum, comparison was carried out among the peaks of blank serum, serum (:on raining drug and Folium Isatidis. decoction to determine the transitional blood components in rats after administration of the de coction. RESULTS After repeated experiments we determine the better method to deal with the serum. Three compounds were obtained,they are medium polar; one of them was found to be high content and had the same retention time with the one in the HPLC chromatogram of decoction,and the similar UV spectrum; other compounds should be metabolites. CONCLUSION The compounds are absorbed into blood and their metabolites are the elective constituents, the original crude drugs are the main in gredients of Folium Isatidis. Its serum pbarmacochemistry should be subiect to complete investigation so as to lay a foundation for the further study of active ingredients of Folium Isatidis.
