摘要
目的研究mucA基因突变对铜绿假单胞菌黏液表型的影响。方法收集临床分离的黏液型铜绿假单胞菌9株,对稳定黏液表型的铜绿假单胞菌采用聚合酶链反应方法对其mucA基因进行扩增,全自动荧光测序仪测序分析。结果筛选出3株稳定的黏液表型的铜绿假单胞菌,编号分别为PA255、PA622和PA880。mucA基因序列分析中,3株均存在第342位(A→G)的无意义突变,PA255第426位G缺失导致第439位产生终止密码,还伴有393位(C→G)的无意义突变,PA880还存在第465位(T→G)的点突变,PA622未发现其它位置的突变。结论mucA基因突变是造成铜绿假单胞菌产生黏液表型的原因之一,同时也存在其它的遗传因素使其产生黏液表型。
Objective To investigate the impact of mucA gene mutation on the mucoid phenotype of pseudomonas aeruginosa. Methods The mucA gene of pseudomonas aeruginosa separated from clinical departments was amplified and the PCR product was sequenced. Results Three stable mucoid phenotype pseudomonas aeruginosa isolates (PA255, PA622, PA880) were screened. All contain a silent A-to-G transition at nucleotide (nt) 342. In addition, the strain PA255 contains the deletion of a G residue at nt 426, which affects the homopolymeric strech of five G residues between positions 425 and 430, accompanying with a silent C-to-G transition at nt 393. The strain PA880 contains a T-to-G transition at nt 465. No mutation at other position is present in the strain PA622. Conclusion The mucA mutation is one important pathway to mucoid phenotype of pseudomonas aeruginosa, and there are other genetic factors that also affect the phenotype of pseudomonas aeruginosa.
