金属鳌合亲和层析填料的制备及其在重组六聚组氨酸融合蛋白中的应用 被引量:3

摘要 目的:制备Ni2+螯合金属层析填料,分离HSP65-MUC1-histag基因工程蛋白质。方法:应用Sepharose4B作为载体,亚氨基二乙酸(IDA)为起始材料合成金属鳌合层析填料,同时应用5-Br-PADAP一阶导数光度法对Ni2+的吸收峰进行鉴定和定量。最后应用制备的Ni2+-Sepharose4B纯化HSP65-MUC1-histag。结果:Ni2+最佳吸收峰为574nm,凝胶中螯合的Ni2+为5.2mg.g-1,Ni2+-Sepharose4B金属螯合亲和层析纯化重组蛋白纯度≥98%,回收率≥90%。结论:制备了能应用于蛋白质亲和层析的高效金属鳌合层析填料。
