摘要 目的:探讨昆明鼠的遗传变异鼠-昆明-无毛(kun ming hairless,KM-HL)鼠对肿瘤细胞的几项生理指标与其他小鼠的区别,为将KM-HL鼠作为肿瘤实验模型用鼠提供理论依据。方法:用TNF-αELISA酶免试剂盒测试KM-HL鼠血清的肿瘤坏死因子(tumornecrosis factor,TNF);将KM-HL鼠接种白血病瘤株(L1210)后,测定TNF-α并计算脾系数、胸腺系数等生理指标;用IL-2ELISA酶免试剂盒测试KM-HL鼠血清的白细胞介素2(interleukin,IL-2);以上两种酶免试剂盒测试方法均用酶标仪在450nm处测定OD值;将KM-HL、KM(昆明种)及ICR鼠接种肝癌H22肿瘤细胞,10d后取肿瘤、肝脏、脾脏及胸腺称质量,计算脾系数和胸腺系数。结果:正常KM-HL鼠的TNF-α为0.144±0.017,比ICR鼠的0.170±0.04低;正常KM-HL鼠接种L1210后血清中的TNF-α为0.176±0.028,比空白KM-HL鼠的0.144±0.017高,P<0.05。KM-HL鼠IL-2为0.137±0.025,与ICR鼠的0.155±0.077基本相似,差异无统计学意义。在肝癌H22肿瘤模型实验中,KM-HL鼠的肿瘤[(2.966±0.94)g]比ICR鼠[(1.262±0.61)g]的肿瘤生长快,差异有统计学意义,P<0.01;KM-HL鼠[(10.461±2.69)mg/g]比KM鼠[(14.959±4.154)mg/g]、ICR鼠[(18.826±4.28)mg/g]的脾系数小,KM-HL鼠[(1.212±0.49)mg/g]比KM鼠[(3.315±0.901)mg/g]、ICR鼠[(2.914±1.28)mg/g]的胸腺系数也小,差异有统计学意义,P值均<0.01。结论:KM-HL鼠对于肿瘤细胞具有易感性,且肿瘤生长均一性好易统计,适合作为肿瘤实验模型用鼠用于抗肿瘤药物的筛选。 OBJECTIVE: To study the differences of KM mice's variation mice-KM-HL mice with other kinds of mice on some physiological indexes of tumor cells in order to provide the theoretical foundations for KM-HL mice used as experimental model mice. METHODS: Tumor necrosis factor (TNF-α) of KM-HL mice was tested by TNF-α ELISA kit. After injecting tumor cells of leukemia Ll210 on KM-HL mice, TNF-α was tested, while some of the physiological indexes were calculated such as spleen index, thymus index etc. Serum interleukin-2 (IL-2) of KM-HL mice was tested by IL-2 ELISA kit; When using both of the two ELISA kit, OD value of 450nm was measured by enzyme immunoassay instrument; Ten days after injecting tumor cells of hepatoma H22 on KM-HL, KM(Kunming mice) and ICR mice, tumors, livers, spleens and thymuses were taken and weighted, and the spleen index and thymus index were calculated. RESULTS: TNF-α of normal KM-HL mice (0. 144±0.017) was lower than that of ICR mice (0. 170±0. 04), but after injecting tumor cells of leukemia L1210, TNF-α of serum (0. 176±0. 028) iu- creased compared with the blank KM-HL mice (0. 144±0. 017), and had a significant difference, P〈0.05. IL-2 of KM-HL mice (0. 137±0. 025) was similar to ICR mice (0. 155±0. 077), and had no any statistic difference. In the hepatoma H22 model experiment, the tumors of KM-HL mice (2. 966±0.94) g grew faster than that of ICR ones (1. 262±0. 61) g, and had a significant difference, P〈 0.01. The spleen index of KM-HL mice (10. 461±2.69) mg/g was smaller than that of KM (14. 959±4. 154) mg/g and that of ICR mice (18. 826 ± 4.28) mg/g, while the thymus index of KM-HL mice (1. 212 ±0.49) mg/g was also smaller than that of KM (3.315±0.901) mg/g, and that of ICR mice (2.914±1.28) mg/g, and have statistical significance, P〈0.01.
