摘要
目的了解引起江苏省2005-2006年麻疹流行的麻疹野病毒基因特征,为制定消除麻疹策略提供依据。方法2005-2006年收集了324例来自江苏省7个设区市的麻疹急性期病人咽拭子标本,用EB病毒转化的狨猴淋巴母细胞(B95a细胞)从5个市分离到麻疹病毒99株。用逆转录-聚合酶链反应从99株麻疹病毒中扩增出核蛋白(N)基因3’端676个核苷酸片段,并对该片段进行序列测定和分析,构建基因亲缘关系树。结果通过N基因3’端450个核苷酸片段的序列测定和分析证明,99株均为麻疹病毒H1基因型中的H1a基因亚型,H1a基因亚型中有2个小分支。99株麻疹病毒间450个核苷酸差异率为0%~4.7%;与A基因型代表株Edmonston株的核苷酸差异率为6.7%~9.8%;与H2基因型代表株China94—1的核苷酸差异率为6.7%~9.6%。结论H1a基因亚型麻疹病毒在江苏省广泛流行,为绝对优势基因亚型,H1a亚型2个小分支中的很多不同或相同的病毒株引起的多个传播链造成省内各市的麻疹广泛传播。
Objective Conducting the genetic characterization of wild-type measles viruses isolated in Jiangsu Province between 2005 to 2006, and to provide scientific evidence for accelerating measles control program and measles elimination in the future in Jiangsu Province. Methods 324 throat swabs were collected from acute measles patients from 7 prefectures of Jiangsu Province between 2005 to 2006. Ninety-nene strains of wild-type measles viruses were isolated using B95a from 5 Prefecture's specimens. Fragment of 676 nucleotide acids of the carboxyl end of nucleoprotein gene were amplified using reverse transcription polymerase chain reaction (RT-PCR) method,and the PCR products were directly sequenced and analyzed. The phylogenetic tree was constructed based on 450 nucleotide acids of the carboxyl end of nucleoprotein gene,and homological analysis was performed at nucleotide acid level. Results The phylogenetic analysis of the isolates compare to WHO reference strains indicated that all of the 99 isolates belonged to H1a sub genotype of H1 genotype. Nucleotide acid variation were 0% to 4.7% among the 99 iolate; 6.7% to 9.8% compare the A genotype(Edmonston strain);6.7% to 9.6% compared with H2 genotype (China 94-1). Conclusion H1a sub-genotype was the major genotype of wild type measles viruses circulated in Jiangsu from 2005 to 2006, H1b and H1c were not found. There were 2 small clusters within H1a sub-genotype virus co-circulating and caused the re-emergency of measles in Jiangsu in 2005 and 2006.
