摘要
目的研究缺氧诱导因子-lα(HIF-lα),Caspase-3,Survivin在食管癌的发生、发展中的作用。方法应用免疫组化SP法检测49例食管鳞癌中HIF-lα、Caspase-3、Survivin蛋白的表达。结果HIF-lα蛋白在食管鳞癌组织中的阳性率为51.0%(25/49),显著高于正常食管黏膜0(0/32)(P<0.05);Caspase-3蛋白在食管鳞癌组织中的阳性率为34.7%(17/49),显著低于癌旁正常黏膜100%(32/32)(P<0.05);Survivin蛋白在食管鳞癌组织中的阳性率为57.1%(28/49),显著高于癌旁正常黏膜6.3%(2/32)(P<0.05)。结论HIF-lα蛋白Survivin蛋白的高表达和Caspase-3蛋白的低表达,在食管鳞癌发生、发展中起重要作用,HIF-lα蛋白的阳性表达可能下调下Caspase-3蛋白的表达,使Survivin蛋白的表达增加。联合检测HIF-1α、Caspase-3与Survivin可作为判断食管癌生物学行为的客观指标,对判断食管癌的预后具有重要意义。
Objective To explore the relationship between the expressions of HIF -1α, Caspase - 3, Survivin proteins and the occurrence and development of esophageal squamous cell carcinoma. Methods The expressions of HIF -1α, Caspase - 3, Survivin protein were detected in 49 eases of esophageal squamous cell carcinoma, and cancer- adjacent mueosa tissues( including normal epithelium, simple hyperplasia epithelium, mild atypical hyperplasia epithelium, severe atypical hyperplasia epithelium, carcinoma in situ) by streptomyeete avidin peroxides(SP) immunohistochemistry, ,Results There were increasing trade in the positive rates of HIF - 1α, Survivin protein from normal epithelium, simple hyperplasia epithelium, mild atypical hyperplasia epithelium, severe atypical hyperplasia epithelium, carcinoma in situ to invasive carcinoma, and there were significant differenees between the normal epithelium and the invasive carcinoma ( P 〈0.05). There were decreasing trade in the positive rates of Caspase - 3 protein from normal epithelium, simple hyperplasia epithelium, mild atypical hyperplasia epithelium, severe atypical hyperplasia epithelium, carcinoma in situ to invasive carcinoma, and there were significant differences between the normal epithelium and the invasive carcinoma ( P 〈 0.05 ). There was a positive correlation between the positive expression rate of HIF - 1α protein and the that of Survivin protein, and a negative correlation between the positive expression rate of HIF -1α protein and the that of Caspase - 3 protein in esophageal squamous cell carcinoma( P 〈 0.05). Conclusion The over expression of HIF -1α, Survivin protein and decrease expression of Caspase - 3 protein can play important roles in the occurrence and development of esophageal squamous cell carcinoma. The expression of HIF -1α protein can decrease the expression of Caspase - 3 protein, accordingly,increasing the expression of Survivin protein.
