摘要
目的:探讨天龙咳喘灵胶囊(TLC)对慢性常压低氧性肺动脉高压大鼠的防治作用。方法:24只SD大鼠随机分为:正常对照组、单纯低氧组和低氧+TLC组,观察肺组织形态学变化,测定各组大鼠平均肺动脉压(mPAP)、体循环平均压(mSAP)和右室肥厚指数(RVH I);计算动脉中膜厚度占外径的百分比(MT%)、管壁面积占血管总面积的百分比(WA%)和管腔面积占血管总面积的百分比(LA%);检测动脉血中是一氧化氮NO含量和一氧化氮合酶NOS活性。结果:(1)与正常对照组相比,低氧组大鼠肺组织切片HE和弹力纤维染色可见肺小动脉壁增厚、管腔狭窄和肺泡隔增厚等改变,TLC干预后可显著减轻低氧所致的肺组织病理改变;低氧组mPAP、RVH I、MT%和WA%显著高于正常对照组(P<0.01),而低氧+TLC组与正常对照组相比差异无统计学意义(P>0.05);低氧组LA%与正常对照组和低氧+TLC组相比显著减小(P<0.01)。3组mSAP差异无统计学意义(P>0.05)。(2)慢性低氧使大鼠动脉血中NO含量和NOS活性显著降低(P<0.01),而低氧+TLC组与正常对照组相比差异无统计学意义(P>0.05)。结论:TLC可抑制大鼠慢性低氧性肺动脉高压的形成,其机制可能与其增强NOS活性、增加NO含量有关。
Objective: To investigate the preventive effect of Tianlongkechuan capsules (TLC) on chronic hypoxic-induced pulmonary hypertension in rats. Methods: Twenty-four Sprague-Dawley rats were randomly divided into Control ( C group), Hypoxia ( H group), and Hypoxia + TLC groups ( HT group). Morphology of pulmonary tissue and blood vessels, mean pulmonary arterial pressure (mPAP), mean systemic artery pressure (mSAP) and right ventricle hypertrophy index (RVHI) were evaluated. Percentages of intimea-media thickness to the external diameter (MT%) ,wall area to total transectional area (WA%) and luminal area to total transectional area (LA%) were calculated. NO concentration and NOS activity in blood serum were measured. Results: (1) Microstructure of pulmonary tissue and pulmonary arteries of H group changed obviously with the development of hypoxia-induced pulmonary vascular structural remodeling, and those changes of HT group were ameliorated. (2) mPAP and RVHI of H group [ ( 19.41 ± 3.68 ) mmHg and ( 31.05 ± 5.42 ) ] were significantly higher than those of C group [ ( 14.28 ± 2.09 ) mmHg and (20.78 ± 1.90) ] and HT group [ ( 15.30 ± 2.64) mmHg and ( 22.92 ± 4.25 ) ] ( P 〈 0.01 ). There was no significant difference in mSAP among the three groups ( P 〉 0.05 ). (3) Plasma NO concentration and NOS activity of H group [ (84.25 ± 13.31 ) μmol/L and ( 16.51 ± 3.89 ) U/L ] were markedly decreased compared with those of C group [ ( 105.59 ± 6.95 ) μmol/L and (28.35 ± 1.80 ) U/L ] and HT group [ ( 116.53 ± 15.19 ) μmol/L and ( 24.03 ± 3.54 ) U/L] (P 〈 0. 01 ). Conclusion: These results indicate that TLC may have a preventive effect on chronic hypoxiainduced pulmonary hypertension, which may be related to enhanced NOS activity and alteration of NO level.
