摘要
目的:建立以反相高效液相色谱法测定宁心红杞胶囊中金丝桃苷和绿原酸含量的方法。方法:色谱柱为Kromasil C18(250mm×4·6mm,5μm),流动相为乙腈-0·1%磷酸(梯度洗脱),流速为1mL·min-1,柱温为30℃,金丝桃苷检测波长为205nm,绿原酸检测波长为325nm。结果:金丝桃苷和绿原酸的检测浓度分别在0·0151~0·2414mg·mL-1(r=0·9993)和0·0126~0·2018mg·mL-1(r=0·9991)范围内与各自峰面积积分值呈良好线性关系;金丝桃苷和绿原酸的平均回收率分别为98·89%和97·95%,RSD分别为0·52%和0·82%。结论:本方法简便、快速、灵敏度高,可用于宁心红杞胶囊的质量控制。
OBJECTIVE : To establish a RP - HPLC method for the content determination of Hyperoside and Chlorogenic acid in Menoprogen capsules. METHODS: The samples were separated on Kromasil C18 column, with mixtures of 0.1% phosphoric acid and acetonitrile as mobile phase in a gradient elution at a flow rate of 1.0 mL · min ^-1. The column temperature was 30 ℃ ;the detection wavelength of Hyperoside was 205 nm and that of Chlorogenic acid was 325 nm.RESULTS: Hyperoside and Chlorogenic acid showed good linear relationship over the ranges of 0.015 1-0.241 4 mg · mL^-1( r = 0.999 3) and 0.012 6-0.201 8 mg · mL^-1(r = 0.999 1), respectively. The average recoveries were 98.89% and 97.95% with RSD at 0.52% and 0.82%, respectively. CONCLUSION: This method is simple, rapid and sensitive, and it can be used for the quality control of Menoprogen capsule.
