摘要
目的探讨单味蛴螬提取物对兔视网膜静脉阻塞RVO模型的干预作用。方法取健康家兔18只,每组6只,分别为健康空白组;模型组;蛴螬提取物组。术后3、7、14 d用空气栓塞法将动物处死后,迅速摘取标本,脱水,石蜡包埋切片,做视网膜的HE染色病理观察和iNOS表达的免疫组化,计算机系统图像分析结果并进行统计分析。结果蛴螬组3、7、14 d与模型组同时相点比较视网膜组织内层神经纤维层,内核层及外核层厚度均增加;3组的平均光密度值和平均灰度值的组间比较,差异均有非常显著统计学意义(P<0.01)。视网膜阳性反应细胞平均光密度值越低,其对应的iNOS表达就越弱;相反平均灰度值越低,其对应的iNOS表达就越强。结论蛴螬提取物可以减轻兔RVO模型后视网膜各层细胞受到的损害,有效保护视网膜视神经细胞。蛴螬提取物可以减弱兔RVO模型后视网膜由于缺氧而诱导的iNOS表达,由此可减轻NO的过量生成对视网膜视神经细胞造成的毒性伤害作用。
Objective To explore the effect of single grub extract on retinal vein occlusion (RVO) in rabbits. Method The 18 rabbits were randomly divided into 3 groups, including sham-operated, model and grub extract model groups. The RVO rabbits were sacrificed after 3, 7 and 14 day respectively with airembolism and the specimens were collected, dehydrolyzed, paraffin embedded and made sections, then, the morphological analyses were performed and the expression of iNOS was detected, all the results were statistic. Result The thicknesses of the retinal nerve fiber layer, the inner nuclear layer and the inner nuclear layer in grub extract group after 3, 7 and 14 days were increased obviously than that in model group; and the differences of average optical density and average gray value in these 3 groups were significant (P〈0.01). The lower average optical density was, the weaker expression was. On the contrary, the lower average gray value was, the stronger expression was. Conclusion Grub extract can alleviate the injuries of the every layers of the retina induced by RVO, and protect effectively retinal optic nerve cells. Grub extract can also weaken the expression of iNOS induced by hypoxic ischemia, which indicates that Grub extract can alleviate the toxic injuries caused by over production NO.
