辐照魔芋葡甘露聚糖的应用研究 被引量:11

摘要 魔芋葡甘露聚糖(KGM)经60Coγ-射线辐照后,用β-甘露聚糖酶水解制备葡甘露低聚糖,然后用活性炭、离子交换树脂纯化葡甘露低聚糖,最后用凝胶渗透色谱(GPC)和质谱(MS)鉴定产物。结果表明,辐照是一种有效的前处理方法,不仅可以显著地降低KGM的黏度,还可以在一定程度上提高其酶解效率。纯化后的葡甘露低聚糖液的总脱色率为89.2%,脱盐率为83.8%,可溶性固形物损失率为15.1%。GPC和MS分析结果显示,辐照魔芋葡甘露聚糖经酶解24h后所得产物为8个葡萄糖或甘露糖组成的低聚糖。 Konjak glucomannan (KGM) was irradiated by ^60Co γ-irradiation and then hydrolyzed by β- mannanase to prepare glucomannan oligosaccharide, then active carbon and ion-exchange resin were used to purify glucomannan oligosaccharide solution and gel permeation chromatography (GPC) and mass spectrum (MS) were used to identify the purified production. Experimental results showed that γ-irradiation was an effective pretreatment method of KGM, not only reduce viscosity method of KGM evidently but also enhance the efficiency of enzylomysis in a certain degree. The total decoloration rate and deminerization rate of glucomannan oligosaccharide solution were 89.2% and 83.8% respectively; meanwhile, 15.1% souble solid content was lost. GPC and MS indicated that the purified production was polymefized by 8 molecule of glucose or mannose.
