纳豆菌高产培养基的成分优化 被引量:6

Optimizing the Ingredient of Medium for the High Yeald of Bacillus Natto

摘要 为获得生长活力较强,能够满足发酵生产纳豆激酶要求的菌种,以OD600为评价指标,采用测定菌种生长曲线的方法优化培养基配比,利用单因素试验确定适宜菌种生长的培养基成分,并以得到的适宜条件作为五元二次旋转正交设计的零水平进行系统优化。试验结果表明:培养基成分的最优组合为葡萄糖3%、大豆蛋白胨2%、MgSO40.2%、K2HPO40.1%、KH2PO40.2%。在优化条件下,菌种培养14h后菌液的OD600值可达到2.540,较优化前提高了0.674。 In order to gain the strain which has high growth vigor and whose biomass concentration can satisfy further fermentation to produce nattokinase, took OD600 as evaluative index, measured the growth curve to optimize the medium, determined optimal ingredient of medium for the growth of strain by single factor experiment, and then the results of single factor experiment were made as zero level of penta basic-second order revolving orthogonal experiment to systematically determine the optimal combination. The results of this experiment showed that the optimal ingredient of medium was glucose 3%, soybean peptone 2%, MgSO4 0.2%, K2HPO4 0.1%, KH2PO4 0.2%. The OD600 of strain culture at 14h could reach 2.540 under optimal condition and the OD600 increased 0.674 after optimization.
