摘要
目的观察异亚丙基莽草酸(ISA)对人宫颈癌Hela细胞转录因子STAT1、STAT3、NF-κB表达作用的影响,探讨其抗炎作用机制。方法用MTT法体外检测ISA不同浓度对Hela细胞生长的抑制作用,采用双荧光素酶报告系统检测不同浓度药物对转录因子STAT1、STAT3、NF-κB表达活性的影响。结果ISA对人宫颈癌Hela细胞的生长没有明显的抑制作用;ISA处理组NF-κB-luc荧光素酶活性下降,而GAS-luc、STAT3-luc荧光素酶活性无明显改变。结论ISA可能通过下调转录因子NF-κB的活性,抑制炎症因子的表达,对炎症反应起到抑制作用。
Objective To study the effect of 3,4-oxo-isopropylidene-shikimic acid (ISA) on transcription factor STAT1, STAT3 and NF-κ B in human cervix cancer Hela cell. Methods The cell proliferation was assessed by MTT assay. The luciferase activity of transcription factor STAT1, STAT3 and NF-κ B was determined by the Dual-Lucfferase Reporter Assay System. Results ISA can not inhibited the growth of Hela cell. The luciferase activity of transcription factor NF-κ B in Hela cells treated with ISA was inhibited, while the luciferase activities of transcription factor STAT1 and STAT3 were not inhibited. Conclusion ISA can inhibit inflammation, which may be related with suppression of NF-κ B transcriptional activity.
